Optimizing Micropropagation Protocol of Rose (Rosa hybrida L.) cv. ‘Double Delight’

An experiment was conducted to develop a suitable in vitro plant regeneration technique for Rosa hybrida L. cv. ‘Double Delight’ using nodal segments and leaf tissues as explants through direct indirect organogenesis. Aseptic were inoculated onto gelled MS medium contained various strengths of growth regulators alone combinations callus shoot induction. containing 2.0 mg/l BAP found be the most effective induction from (87.5%) it produced maximum number shoots per explant. The highest frequency 80% tissue, 72% 96% intermodal raised plantlets 2.5 2, 4-D. best percentage organogenesis (93.33%) with length when different explant-derived transferred 3.0 supplemented medium. indirectly induced multiplied on BAP, where average culture 7.20 ± 0.80 5.22 0.47 cm, respectively. Maximum rooting (80%) observed ½-strength 15.0 g/l sucrose 1.0 IBA. Plantlets proper root system then placed polybag 1:1:1 ratio sand, garden soil, compost, they had survival rate about 76%.
